Evaluation of the optimal culture conditions of red mold rice and lactic acid bacteria

Description

        Traditional culture methods based on biochemical characteristics are labor intensive, and typically require up to 72 h completing. Rapid detection techniques directed at similar immunological and genetic targets are therefore of great interest, especially since it is very difficult to determine the total number of bacteria present in foods or fecal samples. Immunological methods based on the detection of shiga-like toxin have been developed. We provided a duplex TaqMan® real-time PCR method for the detection of E. coli O157 that showed good specificity, sensitivity as well as saving substantial time for the preparation of samples without pre-culture. In the future, we will try to create other methods for isolating the DNA of pathogens and the removal of PCR inhibitors from other different samples which are related to E. coli O157:H7. Shortening the process time and increasing the specificity of the pathogens detected are critical for the safety and sanitation of our food supply.

        Enterohaemorrhagic Escherichia coli O157:H7 is a very frequently talked about pathogen around the globe lately. Outbreaks of this infection have been reported in Australia, Canada, Japan, the United States, and in various countries in European and southern Africa. In the summer of 2001, the first clinical infection by E. coli O157:H7 was reported in Taiwan. In this study, we set up the standard procedures for molecular subtyping, and analyzed the strains collected from Taiwan as well as areas outside the country. For the epidemiological purpose, pulsed-field gel electrophoresis (PFGE), amplified fragment length polymorphism (AFLP), and random amplified polymorphic DNA (RAPD) methods were used to investigate the collected strains. The isolates from USA, Canada, Japan, and Taiwan have unique molecular subtyping patterns which are according to its distributions of areas. The environmental strains isolated from Taiwan had more relationships and their similarity was about 75-85%. The first clinical strain isolated from Taiwan in 2001 was similar with strains from North American and actually confirmed that this clinical infection was not relative with the local environmental strains. Our surveys showed that E. coli O157:H7 strains actually survive in environment of Taiwan, but there was no official report about the infection by E. coli O157:H7. The diets and geographic distributions were important factors. The establishment and joining of a global network database is an important task if we wish to control the agricultural and food-borne pathogens and reduce the losses due to infection and the spread of the pathogens.

        A nested PCR method has been developed to distinguish traditional soybean from transgenic Roundup Ready soybeans (RRS). Comparing with the standard PCR method, the nested PCR method increases the sensitivity by 200 to 1000-fold. One copy of RRS genomic DNA can be detected by the nested PCR. Sufu, a traditional fermented soybean curd, is a very important Chinese condiment and it is a highly processed food. We have used the nested PCR to detect seven time points of sufu throughout the aging period. The nested PCR method showed positive results for sufu made up of 100% RRS at all seven time points from 0th day to 180th day. This work describes the development and application of a sensitive and specific detection method for sufu made from RRS using a nested PCR system.

 

 

       A milk - soymilk mixture was fermented using Lactobacillus paracasei subsp. paracasei NTU101 and Bifidobacterium longum BCRC11847 at different inoculum ratios (1:1, 1:2, 1:5, 2:1, and 5:1). When the inoculum ratio was 1:2, the cell numbers of both strains were balanced after 12 h of cultivation. The pH and titratable acidity were very similar at the various inoculum ratios of cultivation. The milk–soymilk mixture was supplemented with 5, 10, 15, and 20% Lycium chinense Miller juice and fermented with Lactobacillus paracasei subsp. paracasei NTU101 and B. longum BCRC11847. Sensory evaluation results showed that supplementation with 5% Lycium chinense Miller juice improved the acceptability of the fermented milk–soymilk. The fermented beverage was stored at 4_C for 14 days; variations in pH and titratable acidity were slight. The cell numbers of L. paracasei subsp. paracasei NTU101 and B. longum BCRC11847 in the fermented beverage were maintained at 1.2·109 CFU/ml and 6.3·108 CFU/ml, respectively, after 14 days of storage.

Important Results

Applied Science

         Monacolin K is a secondary metabolite of Monascus species, and has effect on reducing cholesterol level. The objective of this study was to investigate the effect of red mold rice supplementation with different concentration on egg quality, laying performance, serum cholesterol, serum triglyceride, HDL, LDL, serum lipid peroxidation and egg yolk cholesterol in Hy-line laying hens. The research studied 48 Hy-line laying hens of 48 weeks old by dividing them into 4 groups. Except for the control group, the feed for three other groups contained 2.0%, 5.0% and 8.0% red mold rice (monacolin K concentration is 0.0145%, 0.035% and 0.056%). The experiment lasted 6 weeks. During this period, egg weight and egg production were recorded everyday, and cholesterol concentration, triglyceride, HDL, and LDL in serum were measured weekly as well. The result showed that cholesterol level in eggs produced by experimental groups was lower than that of control group (0%: 194.14±8.30, 2%: 167.17±4.34, 5%: 168.93±9.38, 8%: 183.02±7.63 mg/egg; p<0.05), the triglyceride (0%: 1494±178, 2%: 1280±174, 5%: 1189±248, 8%: 1381±218 mg/dL; p<0.05) and LDL level (0%: 36.81±5.53, 2%: 32.25±7.93, 5%: 30.06±4.39, 8%: 28.81±4.16 mg/dL; p<0.05) were also significantly lowered in the experimental groups. However, HDL level did not show significant change for both control and experimental groups (0%: 36.06±3.96, 2%: 36.25±5.39, 5%: 33.13±3.68, 8%: 31.44±4.29 mg/dL; p>0.05). Besides, adding red mold rice also helps on inhibiting production of malondialdehyde (MDA) in serum lipid oxidation (0%: 27.42±0.53, 2%: 25.62±0.76, 5%: 24.35±0.59, 8%: 23.63±0.48 μM; p<0.05). HPLC analysis showed that no monacolin K residues were detected in egg yolk and albumen extracts from hens fed red mold rice.

 
 

Yolk color score showed that the darkest color happened when 8.0% of red mold rice was incorporated into the feed and the lightest yolk color was in the control group.
